FBLN1

Fibulin-1 (encoded by FBLN1), also known as BM-90, is an extracellular matrix (ECM) glycoprotein critical for ECM assembly and structural integrity. It binds collagen IV, elastin, and laminin, regulating cell adhesion, migration, and tissue remodeling. It also modulates TGF-β signaling by controlling ligand bioavailability, influencing cell proliferation and differentiation.
Dysregulated Fibulin-1 links to aortic aneurysms (reduced levels impair ECM stability) and cancer metastasis (overexpression enhances ECM remodeling in breast/lung cancers). Preclinical studies explore targeting its pathways: monoclonal antibodies against functional domains and small molecules inhibiting ECM interactions, as potential therapies for these disorders.
