GSDMA is a member of the Gasdermin family and is mainly associated with the process of pyroptosis. It consists of several exons and encodes a protein with cytoplasmic localization that can cause cell membrane perforation after activation. The function of GSDMA involves the inflammasome pathway, in which the activation of the inflammasome leads to the activation of caspase-1, which then cleaves GSDMA, releasing its N-terminal domain and triggering pyroptosis. In addition, the activation of GSDMA is also associated with caspase-3, which can promote the process of pyroptosis. GSDMA also plays a role in tumor development and therapeutic response, and its activation may inhibit tumor cell proliferation and shrink tumors. Immunofluorescence staining of HepG2 cell with CSB-PA850415LA01HU at 1:30, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and and permeated by 0.2% TritonX-100 for 15 min. Then 10% normal goat serum to block non-specific protein-protein interactions . The cells were then incubated with the antibody overnight at 4℃. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
