Interleukin-associated immunoglobulin-like receptor-1 (LAIR1) is a transmembrane glycoprotein containing an Ig-like extracellular domain and a member of the immunoglobulin superfamily. It can be expressed on various immune cell membranes, and can also be shed from the cell membrane surface to become soluble LAIR1 (sLAIR1) in serum. LAIR1, also known as CD305, is a class of inhibitory receptors that were successfully cloned in 1997 and can exert its immunosuppressive function through the immunoreceptor tyrosine inhibitory motif (ITIM) in its cytoplasm.
Human LAIR1 is a type I transmembrane protein with a leader sequence consisting of 21 aa, 142aa in the extracellular region and 101aa in the cytoplasmic region. The cytoplasmic region of LAIR1 contains two sequences VTYAQL and ITYAAV, which conform to the ITIM conserved sequence I/V/L/SxYxxL/V, and there are 24 aa between them. When LAIR1 on the cell membrane undergoes tyrosine phosphorylation under stimulation, LAIR1 at this time will recruit SH1 and SH2-containing tyrosine phosphatase (SHP) to transmit obvious inhibitory signals, thereby downregulating NK cell-mediated cells Toxicity, cytotoxicity of cytotoxic T lymphocytes, and TCR-mediated cytokine secretion during T cell activation. At present, many studies have confirmed that the abnormal expression of LAIR-1 is related to the pathogenesis of systemic lupus erythematosus (SLE), rheumatoid arthritis (RA) and other immune diseases.
In addition, Tomasz et al. found that the expression level of LAIR1 molecule on CD4+T cells and CD8+T cells in patients with lung cancer and colon cancer was significantly higher than that in normal people, which could reduce the antitumor response. The study also suggests that the immune escape phenomenon of tumor cells may be related to the role of the inhibitory receptor LAIR1.
