MKRN1

MKRN1 (Makorin Ring Finger Protein 1), alias RNF61, is an E3 ubiquitin ligase mediating target protein ubiquitination. It negatively regulates tumor suppressor p53 by promoting its proteasomal degradation, modulating cell cycle arrest and apoptosis. Additionally, MKRN1 binds mRNA to influence translation and stability, linking it to post-transcriptional gene regulation.
Dysregulated MKRN1 contributes to cancers (breast, colorectal) via reduced p53 levels. Its localization in the 15q11-q13 imprinted region implicates it in Prader-Willi syndrome. Preclinical studies test MKRN1 E3 ligase inhibitors to stabilize p53 and suppress tumor growth, though no clinical trials are ongoing.

IHC image of CSB-PA883390LA01HU diluted at 1:600 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
