PRB2, also known as proline-rich protein BstNI subfamily 2, is a basic, proline-rich salivary glycoprotein. It undergoes proteolytic processing to generate mature isoforms before secretion. PRB2 functions as a tumor suppressor, negatively regulating cell cycle progression and apoptosis. It is involved in signaling pathways such as the p53-p21 DNA damage response pathway. Diseases associated with PRB2 include actinobacillosis and acute vascular insufficiency of the intestine. Drug development for PRB2 focuses on targeted protein degradation (TPD) strategies, utilizing PROTACs and molecular glues to degrade disease-causing proteins.
