PREX2

PREX2 encodes Phosphatidylinositol-3,4,5-trisphosphate-Dependent Rac Exchange Factor 2, a guanine nucleotide exchange factor (GEF) that activates Rac GTPases by catalyzing GDP-GTP exchange. It regulates actin cytoskeleton remodeling, cell migration, and proliferation through interactions with phosphoinositides and signaling proteins like PTEN.
Aberrant PREX2 activity is linked to cancer progression, particularly melanoma and prostate cancer, via PTEN inactivation and PI3K/Akt pathway dysregulation. Preclinical studies investigate small-molecule inhibitors targeting its GEF domain to disrupt oncogenic signaling, though no clinical candidates have advanced to trials.
