RAB27A encodes a small GTPase of the RAS oncogene family, with aliases including ASH1 and melanophilin-binding protein. It regulates vesicle trafficking by cycling between GTP-bound active and GDP-bound inactive states. Active RAB27A interacts with effectors like melanophilin (MLPH) and myosin Va (MYO5A) to mediate transport of lysosome-related organelles (e.g., melanosomes, cytotoxic granules) to the plasma membrane, facilitating exocytosis.
Mutations in RAB27A cause Griscelli syndrome type 2 (GS2), an autosomal recessive disorder marked by partial albinism and immunodeficiency (impaired cytotoxic T/NK cell function). Overexpression is linked to melanoma (promotes metastasis via exosome secretion) and breast cancer (enhances invasion). Preclinical studies target RAB27A: small molecules inhibiting its effector interactions or GTPase activity show anti-tumor promise; gene therapy for GS2 remains in early stages with no approved therapies to date.

Overlay Peak curve showing PC3 cells stained with CSB-RA285197A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
