RORA

RAR-related orphan receptor alpha (RORA), commonly known as RORα, is a member of the nuclear receptor superfamily. As a transcription factor, it binds to ROR response elements (ROREs) in target gene promoters, regulating circadian rhythm, lipid metabolism, and immune homeostasis. It intersects with pathways like NF-κB and PPARγ, balancing pro-inflammatory and metabolic signaling.
Dysregulation of RORA links to rheumatoid arthritis, Alzheimer’s disease, and metabolic syndrome. Preclinical studies have explored RORA agonists—synthetic small molecules and natural compounds like curcumin—for treating these conditions. Though no FDA-approved drugs target RORA yet, ongoing research aims to validate its therapeutic potential in clinical settings.
