SAGE1 (Sarcoma Antigen 1), a tumor-testis antigen gene, typically exhibits a higher expression level in various tumor tissues compared to adjacent non-cancerous tissues, closely correlating with cell proliferation. By competing with INTS6 (Integrator 6) for INTS3 (Integrator 3), SAGE1 disrupts the "brake" mechanism that hinders RNA polymerase II. The newly formed INTS3-SAGE1 complex then binds to the promoter-proximal regions of gene targets, activating gene transcription in cancer-related pathways. This process ultimately leads to cell proliferation, promoting tumor growth and spread.
