Sperm autoantigenic protein 17 (SPA17), also known as sperm surface protein Sp17 or cancer/testis antigen CT70, is a conserved glycoprotein originally identified in reproductive tissues. It functions in sperm-egg interaction by binding zona pellucida glycoprotein ZP3 during fertilization, mediated through its conserved N-terminal heparin-binding domain. Beyond reproduction, SPA17 aberrantly activates MAPK/ERK and PI3K/AKT pathways in somatic cells, promoting proliferation and migration. This ectopic expression correlates with poor prognosis in multiple cancers, including ovarian carcinoma, hepatocellular carcinoma, and multiple myeloma, where it serves as a tumor-associated antigen. Current therapeutic strategies focus on SPA17-targeted immunotherapy, with DNA vaccines and chimeric antigen receptor (CAR) T-cell constructs showing preclinical efficacy in myeloma models. Paradoxically, autoantibodies against SPA17 are implicated in immune-mediated infertility, highlighting its dual role in physiological and pathological contexts. Recent structural studies revealed its hexameric assembly, providing insights for selective inhibitor design.

IHC image of CSB-PA12124A0Rb diluted at 1:200 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
