TIA1

The TIA1 protein, full name TIA1 cytotoxic granule-associated RNA-binding protein (alias TIA-1), is a key RNA-binding molecule. It mediates pre-mRNA alternative splicing and, under cellular stress (e.g., ER stress), binds mRNAs to form stress granules—pausing translation to protect cells from damage. It also contributes to cytotoxic T cell-induced apoptosis via granule exocytosis.
TIA1 interacts with the PERK-eIF2α stress signaling pathway, where eIF2α phosphorylation regulates its activity. Dysregulated TIA1 is linked to neurodegenerative disorders like ALS, driven by abnormal stress granule accumulation. Preclinical research explores small molecules targeting TIA1-related stress granule dynamics for ALS therapy, though no drugs are yet approved.
