TRIM11

Tripartite motif-containing protein 11 (TRIM11), also known as RING finger protein 167 (RNF167), is an E3 ubiquitin ligase harboring a conserved RING domain. It regulates protein stability via ubiquitination—for example, targeting tumor suppressor p53 for degradation—and participates in p53 signaling and Beclin1-mediated autophagy pathways, influencing cell proliferation and survival.
Overexpression of TRIM11 correlates with cancers like glioblastoma, breast, and lung cancer, driving tumorigenesis through p53 inactivation and enhanced invasion. It may also contribute to Alzheimer’s disease by modulating amyloid-beta metabolism. Currently, TRIM11-targeted therapies remain preclinical: small molecules inhibiting its E3 ligase activity and RNA interference strategies are under investigation for anti-tumor applications, with no approved drugs yet.
