USP28, ubiquitin-specific peptidase 28, alias USP21.This is a deubiquitinating enzyme expressed in human cells and belongs to the USP subfamily of DUB.
USP28 plays a role in the DNA damage pathway and DNA damage-induced apoptosis, and its overexpression has been observed in a variety of cancers including gliomas and non-small-cell lung cancers. USP28's mechanism of action is to regulate protein stability and function by removing ubiquitination modifications on target proteins. It stabilizes oncoproteins and promotes cancer development, but some recent studies have also revealed the oncogenic role of USP28 in certain cancers, such as melanoma.
USP28 may have a dual role in cancer therapy, acting as a target to promote cancer development, but also being involved in mechanisms of cancer inhibition.
