VGLL3, degenerate-like family member 3. also known as VGL3, VGL-3. This is a transcriptional cofactor expressed in humans and belongs to the Vestigial-Like family.
The mechanism of action of VGLL3 involves binding to the TEA structural domain (TEAD) transcription factor and is involved in a variety of biological processes, including musculogenesis, cellular proliferation, and gender-biased autoimmune disorders. VGLL3 is specifically induced in cardiac and hepatic fibroblasts and promotes the collagen production during fibrogenesis. VGLL3 is translocated to the nucleus via matrix stiffness and undergoes liquid-liquid phase separation (LLPS), which promotes collagen production.VGLL3 is also involved in the DNA damage response (DDR), where it is recruited to DNA damage sites in a PARylation-dependent manner, affecting the cellular sensitivity to chemotherapeutic agents.
The biological significance of VGLL3 lies in its role in cardiac fibrosis, tumor development, and DNA double-strand break repair, and is a potential target for fibrosis and tumor therapy.
