WNT6

WNT6 (Wnt family member 6) is a secreted glycoprotein of the Wnt ligand family, commonly aliased as WNT-6. It activates the canonical Wnt/β-catenin pathway by binding to Frizzled (Fz) receptors and LRP5/6 co-receptors, stabilizing β-catenin to enable nuclear translocation and regulate genes involved in cell proliferation, differentiation, and tissue homeostasis.
Dysregulated WNT6 expression links to breast, colorectal, and cutaneous cancers, promoting tumor growth and metastasis. It also contributes to developmental defects like limb anomalies. Drug development focuses on Wnt pathway components (e.g., Fz antagonists, β-catenin inhibitors) for WNT6-associated malignancies, though specific WNT6-targeted therapies remain in preclinical stages.
