The protein encoded by AMFR is Autocrine Motility Factor Receptor, commonly known as GP78. As an E3 ubiquitin ligase, it mediates endoplasmic reticulum-associated degradation (ERAD), regulating misfolded protein clearance. It also modulates pathways like cholesterol homeostasis and cell migration via ubiquitination of target proteins.
AMFR is linked to aggressive tumors (e.g., liver and breast cancer) due to its role in enhancing cell motility and invasion. Preclinical studies are exploring small-molecule inhibitors targeting its E3 ligase activity to suppress tumor progression, though no candidates have entered clinical trials yet. It may also be implicated in neurodegenerative disorders via ERAD dysfunction.

IHC image of CSB-PA333164LA01HU diluted at 1:100 and staining in paraffin-embedded human small intestine tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
