Aspartate β-hydroxylase (ASPH), also known as β-hydroxylase or HAAH, catalyzes the post-translational β-hydroxylation of aspartyl and asparaginyl residues in epidermal growth factor (EGF)-like domains of proteins. This enzymatic modification critically regulates Notch, Jagged, and other EGF-domain-containing receptors, potentiating their activation in tumorigenic signaling pathways. ASPH overexpression drives malignant progression via Notch, Hedgehog, and TGF-β pathways, promoting angiogenesis, epithelial-mesenchymal transition, and immunosuppression within tumor microenvironments. Clinically, ASPH upregulation correlates with aggressive phenotypes in hepatocellular carcinoma, pancreatic ductal adenocarcinoma, and triple-negative breast cancer, serving as a prognostic biomarker. Therapeutic targeting of ASPH has advanced through small-molecule inhibitors (e.g., MO-I-1150) that block its hydroxylase domain and suppress tumor growth in preclinical models. Immunofluorescence staining of Hela cells with CSB-PA002226HA01HU at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C.
