The ATP2A2 gene encodes sarco/endoplasmic reticulum Ca²⁺-ATPase 2 (SERCA2), a P-type ATPase also known as calcium pump 2. It catalyzes ATP-dependent Ca²⁺ translocation from the cytoplasm into the endoplasmic reticulum (ER) or sarcoplasmic reticulum (SR) lumen, maintaining intracellular Ca²⁺ homeostasis critical for muscle contraction (SR in cardiac/skeletal muscle) and cell signaling (ER in non-muscle cells).
Mutations in ATP2A2 cause Hailey-Hailey disease, a chronic skin disorder characterized by acantholysis due to impaired keratinocyte Ca²⁺ regulation. The cardiac isoform SERCA2a is a therapeutic target for heart failure; istaroxime, a SERCA2a activator, is under clinical evaluation for acute decompensated heart failure, enhancing cardiac contractility via increased SR Ca²⁺ uptake. SERCA2 also participates in ER stress and autophagy pathways.

IHC image of CSB-PA002422LA01HU diluted at 1:50 and staining in paraffin-embedded human prostate tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. Secondary antibody only control: uses 1% BSA instead of primary antibody
