BARD1

BARD1, BRCA1-related RING domain protein 1, alias BRCC3, BARD-1, BRCC45, BRCC, is an E3 ubiquitin ligase that forms a complex with the BRCA1 protein and participates in the process of repairing DNA double-strand breaks.
The mechanism of action of BARD1 involves promotion of DNA end resection and protection of DNA replication forks from non planned degradation.The BARD1-BRCA1 complex directly promotes the long-range DNA end resection pathway catalyzed by EXO1 or DNA2 nuclease and stimulates the DNA unwinding activity of Werner or Bloom deconjugating enzymes in the DNA2-dependent pathway. The BARD1-BRCA1 complex also interacts with MRE11-RAD50-NBS1 and phosphorylated CtIP to form the BRCA1-C complex, which synergistically stimulates resection.
BARD1 is essential for maintaining genomic stability and preventing cancer development, and its aberrant function has been associated with the development of several cancers.
