BCAM (Basal Cell Adhesion Molecule), also known as Lutheran glycoprotein (Lu) or CD239, is an immunoglobulin superfamily transmembrane receptor. It binds laminin α5 in the extracellular matrix to mediate cell adhesion, migration, and mechanotransduction. BCAM is functionally linked to the PI3K/AKT and ERK/MAPK signaling pathways, influencing cytoskeletal reorganization and cellular responses to shear stress. Clinically, BCAM is implicated in sickle cell disease by promoting vaso-occlusion through erythrocyte-endothelial interactions. Its overexpression in epithelial cancers (e.g., colorectal, ovarian) correlates with metastasis and poor prognosis. Therapeutic targeting of BCAM-laminin interactions using monoclonal antibodies (e.g., BCAM-05) is under preclinical investigation for sickle cell disease, while small-molecule inhibitors are being explored in oncology to disrupt BCAM-mediated tumor dissemination.

Immunofluorescence staining of HepG2 cells with CSB-PA341920LA01HU at 1:200, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
