BMP1

Bone Morphogenetic Protein 1 (BMP1) is a zinc-dependent metalloproteinase of the astacin family, commonly aliased as procollagen C-proteinase (PCP). It mediates extracellular matrix (ECM) maturation by cleaving C-terminal propeptides of type I/II/III procollagens, enabling formation of functional collagen fibrils critical for tissue structure and integrity.
BMP1 crosstalks with TGF-β signaling pathways to regulate ECM homeostasis. Its dysregulation links to connective tissue disorders like Ehlers-Danlos syndrome and bone developmental anomalies. Drug research focuses on BMP1 inhibitors for fibrotic diseases (e.g., liver fibrosis) where collagen overaccumulates, with preclinical studies showing promise but no approved therapies to date.
