This subunit forms the ion-conducting pore, enabling voltage-dependent calcium influx into cells. It is predominantly expressed in neuronal tissues (e.g., brainstem, inner ear), pancreatic beta cells, and cardiac pacemaker cells, where it modulates cellular excitability and signaling.
Mutations in CACNA1D are linked to Timothy syndrome type 2 (characterized by cardiac arrhythmias and developmental delays) and autism spectrum disorders. Pharmacologically, Cav1.3 inhibitors are being explored for treating hypertension and neurodegenerative conditions, while activators may enhance insulin secretion for diabetes management—though clinical applications remain in early stages.
