CD226 is an important adhesion molecule that belongs to the immunoglobulin superfamily. CD226, also known as DNAM-1, is widely expressed on the surface of a variety of immune cells, including NK cells, T cells, NKT cells, platelet lineage, monocytogenes, activated vascular endothelial cells, and various transformed hematopoietic cell lines. CD226 is involved in important immune processes such as platelet activation and aggregation, killing of CTL and NK cells, and endothelial cell adhesion, and are associated with many important diseases. However, CD226 is not expressed in granulocytes and erythrocytes, in addition, CD226 is not expressed in certain tumor cell lines, such as fibroblasts and epithelial, neural-derived tumor cell lines, etc.
CD226 ligands mainly include CD155 and CD112 that are highly expressed on the surface of various tumor cells. Human CD155, also known as PVR, belongs to the IgSF type I transmembrane glycoprotein, which is both a polio-specific receptor and Up-regulated expression in various tumor tissues. Human CD112, also known as nectin or PRR2, was successfully cloned in 1995. It is the receptor of herpes simplex virus and belongs to the nectin family of IgSF. CD112 is highly expressed in CD34+ hematopoietic precursor cells, CD33+ and CD14+ myelomonocytic cell lines, and at low levels in megakaryocyte lineages. Studies have shown that CD112 can mediate cell-to-cell adhesion through the binding of homologous molecules, thereby participating in the regulation of hematopoietic cell/endothelial cell interactions. CD226 interacts with The binding of receptor immunoglobulin-like structure promotes the adhesion of NK cells and tumor cells to enhance the killing activity of tumor cells.
