The protein encoded by CISD1 is CDGSH iron sulfur domain-containing protein 1, often referred to as Miner1. A mitochondrial outer membrane-localized iron-sulfur protein, it maintains cellular iron homeostasis and supports mitochondrial function. Through binding iron-sulfur clusters, it regulates reactive oxygen species (ROS) levels and mitigates oxidative stress, interacting with the Nrf2 antioxidant signaling pathway.
CISD1 dysregulation correlates with neurodegenerative diseases (e.g., Parkinson’s disease, linked to abnormal iron accumulation) and metabolic syndrome. Preclinical studies are exploring small molecules targeting its iron-binding activity as potential treatments for oxidative stress-related disorders, though no CISD1-specific drugs have entered clinical trials to date.
