DRAM1

DRAM1 encodes Damage-Regulated Autophagy Modulator 1, a lysosomal membrane protein involved in macroautophagy induction. It interacts with BECN1 to promote phagophore formation, regulating autophagic flux in response to cellular stress. DRAM1 is linked to the PI3K/AKT/mTOR pathway, where its upregulation antagonizes mTOR-mediated autophagy suppression.
Aberrant DRAM1 expression correlates with cancer progression, particularly in colorectal and breast cancers, where reduced levels associate with chemotherapy resistance. Emerging studies suggest DRAM1 agonists may enhance autophagy-dependent tumor cell death, though clinical development remains preclinical. Its role in neurodegenerative diseases like Alzheimer’s is also under investigation, via modulation of amyloid-beta clearance.
