DRD4, also known as the dopamine D4 receptor, and by other names such as D4 dopamine receptor, D(2C) dopamine receptor, or Dopamine D4 receptor, is a type of G protein-coupled receptor (GPCR) and a member of the dopamine receptor family. It is encoded by the human gene DRD4. DRD4 is expressed in the limbic system of the brain, particularly in regions associated with emotion and complex behavior regulation. Its mechanism of action involves binding to dopamine and other ligands, activating G proteins, and inhibiting adenylate cyclase activity, thereby influencing intracellular signaling pathways.
DRD4 is involved in regulating emotions, cognitive function, attention, and addictive behaviors. Studies have shown that polymorphisms in DRD4 are associated with various psychological and neurodevelopmental disorders, such as attention deficit hyperactivity disorder (ADHD), schizophrenia, obesity, and nicotine addiction. Additionally, the activity of DRD4 is linked to circadian rhythm regulation; it influences the circadian rhythm of contrast sensitivity by modulating the rhythmic expression of NPAS2 in retinal ganglion cells. Therefore, DRD4 is a potential target in neuroscience research and the treatment of related diseases.
