GPR158, G protein-coupled receptor 158, alias mGlyR, is a receptor that is highly expressed in the brain and belongs to the orphan G protein-coupled receptor family.The mechanism of action of GPR158 is unique in that it inhibits G protein signaling by binding to glycine and activating RGS proteins (regulators of G protein signaling), rather than activating G proteins as most GPCRs do. This inhibition controls neuronal activity by regulating second messenger homeostasis, with significant effects on brain physiology.
Abnormal expression of GPR158 has been associated with a variety of disorders, including depression, Alzheimer's disease, glaucoma, and glioma. Activation of GPR158 enhances the activity of RGS proteins, and the two proteins work together to regulate the in vivo balance of second messengers. GPR158 is a potential new target for antidepressant development.

IHC image of CSB-PA735975LA01HU diluted at 1:500 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of U251 cells with CSB-PA735975LA01HU at 1:166, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
