GPR75, G protein-coupled receptor 75", alias G protein-coupled receptor, Probable G-protein coupled receptor 75.GPR75 is a G protein-coupled receptor expressed in the brain and CNS that can be activated by the chemokine CCL5/RANTES and may be coupled with the Heterotrimeric Gq proteins are coupled, and activation promotes inositol trisphosphate production and calcium ion mobilization.GPR75, together with CCL5/RANTES, may play a role in neuronal survival through activation of downstream signaling pathways, including PI3, Akt, and MAP kinases. In addition, CCL5/RANTES may also regulate insulin secretion in pancreatic islet cells through activation of GPR75 .

The biological significance of GPR75 focuses on its potential role in the regulation of insulin secretion and obesity. Studies have shown that GPR75 may play a role in regulating these physiological processes and is associated with body mass index (BMI). For example, one study found that inactive copies of GPR75 were associated with weight loss and reduced risk of obesity. In addition, loss-of-function mutations in GPR75 in mice resulted in reduced weight gain and improved glucose tolerance and insulin sensitivity, suggesting that GPR75 may play an important role in energy homeostasis and metabolism .
