HCAR1, also known as GPR81, is a G-protein coupled receptor (GPCR) that acts as a receptor for the glycolytic product 2-hydroxypropionate (lactate). It is coupled to Gαi, reducing cellular cAMP levels and modulating downstream Gβγ signaling. HCAR1 is involved in various cellular processes, including lipolysis inhibition, neural activities, angiogenesis, inflammation, vision, cardiovascular function, and stem cell proliferation. It also plays a role in cancer pathogenesis and immune regulation.
Recent research has highlighted HCAR1's involvement in epilepsy, where it modulates neuronal activity and seizure thresholds. This has sparked interest in developing novel antiepileptic drugs targeting HCAR1.
