HSPA5, heat shock protein family A member 5", alias BiP, GRP78. it is a molecular chaperone protein that functions mainly in the endoplasmic reticulum and is encoded by the human cell's own genes. the mechanism of action of HSPA5 involves assisting in the proper folding, assembly, and transport of proteins. When cells are subjected to various stress conditions, such as heat shock, oxidative stress, or nutrient deficiencies, the expression of HSPA5 increases significantly in response to protein misfolding and aggregation.HSPA5 binds to unfolded or misfolded proteins by forming a complex that prevents them from aggregating and facilitates their correct folding or refolding.
The biological significance of HSPA5 is mainly reflected in its role in the cellular stress response and the maintenance of protein homeostasis. As a key molecular chaperone of the endoplasmic reticulum stress response, HSPA5 is involved in the unfolded protein response (UPR), which plays an important role in cellular adaptation and survival.HSPA5 has also been implicated in a variety of cellular processes including cell signaling, cell cycle control, apoptosis, and immune response. In tumor cells, high expression of HSPA5 is associated with tumor cell proliferation, invasion, and chemoresistance, making it a potential target in cancer therapy. IHC image of CSB-RA847116A0HU diluted at 1:100 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
