Interleukin 18 receptor accessory protein (IL18RAP) is an indispensable subunit for the ability of the IL-18 receptor (IL18R) complex to mediate high-affinity IL-18 binding and signal transduction. IL18 is a pro-inflammatory cytokine, also known as an inducer of interferon-γ (IFN-γ). Its receptor (IL-18R) consists of two subunits belonging to the interleukin 1 receptor (IL-1R) family. The alpha chain (IL-18Rα or IL18R1) has a weaker binding affinity for IL-18. After IL-18 binds to IL-18Rα, another subunit—IL18RAP (also known as IL-18Rβ)—is subsequently recruited to form a heterotrimeric complex for high-affinity binding. IL-18 stimulation then recruits the myeloid differentiation primary response protein MyD88 through the toll-interleukin-1 receptor (TIR) domain of IL-18R to initiate cell signaling, thereby activating enhancers of NF-κB and MAPK pathways.
