ITAM

ITAM stands for Immune Tyrosine-based Activation Motif. It is not a gene name but a conserved sequence found in the cytoplasmic tails of many immune cell receptors. The ITAM-containing proteins play a crucial role in immune cell activation. When the associated receptor binds to its ligand, the tyrosine residues within the ITAM are phosphorylated, which recruits kinases and initiates downstream signaling cascades, mainly through the PI3K and MAPK pathways.
Dysregulation of ITAM signaling is associated with various autoimmune diseases and immune deficiencies. In drug development, targeting ITAM-related pathways is an emerging strategy. Some small - molecule inhibitors are being investigated to modulate immune responses, aiming to treat diseases where ITAM signaling is aberrant.
