It interacts with ADAM metalloproteinases (e.g., ADAM22) to regulate neuronal cell adhesion and synaptic plasticity, supporting synaptic structure and function in the central nervous system.
Dysregulation of LGI3 may contribute to neurological disorders like epilepsy and neuroinflammation, though research remains preliminary. Preclinical studies explore targeting its ADAM interactions to modulate synaptic signaling, positioning LGI3 as a potential therapeutic target for synaptic dysfunction-related conditions, with no approved drugs currently available.
