MAGEA4 is a member of the MAGEA gene family, members of which encode proteins with 50% to 80% sequence identity to each other. The promoter and first exon of the MAGEA gene exhibit considerable variability, suggesting that the presence of this gene family enables the expression of the same function under different transcriptional control. MAGEA is one of the four major subfamilies of the melanin antigen gene (MAGE) family, the other three subfamilies are MAGEB, MAGEC and MAGED. The tumor rejection antigen encoded by the MAGE gene is processed in cells to produce antigenic peptides, which combine with HLA class I molecules to form complexes, which can be recognized by autologous cytotoxic T cells and induce specific killing of corresponding tumor cells.
Current studies have shown that the MAGEA gene is up-regulated in different types of cancer, such as lymphocytic leukemia, lung cancer, ovarian cancer, melanoma and other solid malignancies. MACEA4 acts as a master regulator of Tregs development and function. Tregs are a subset of T cells that are immunosuppressive, preventing the body from overreacting to self-antigens. In addition, Tregs are involved in tumor escape from the immune system. The number of MAGEA4+ Tregs was increased in peripheral blood and tumor tissue of patients with breast, gastric, liver and some other cancers. Previous studies have shown that the number of MAGEA4+ Tregs is closely related to tumor progression and prognosis of lung cancer patients. Recent studies have reported that MAGEA4 is not only expressed in Tregs, but also in tumor cells, and MAGEA4 expression in different tumor cells can play different roles.
