MDFIC2

MDFIC2 (MyoD Family Inhibitor Domain Containing 2) encodes a protein with a conserved inhibitor domain that targets MyoD family transcription factors (e.g., MYOD1, MYOG). It negatively regulates myogenic differentiation by blocking the DNA-binding activity of these factors, thereby suppressing muscle cell maturation and lineage commitment.
This protein intersects with myogenic signaling pathways including Wnt/β-catenin and TGF-β. Dysregulation of MDFIC2 is linked to muscular dystrophies (impaired muscle regeneration) and rhabdomyosarcoma (aberrant myogenic cell proliferation). Preclinical studies are exploring small molecules to disrupt its inhibitory interaction with MyoD factors, with potential applications in muscle repair and anti-tumor therapy.
