Mitofusin-2 (MFN2), alias MARF or HMSN6, is a mitochondrial outer membrane GTPase essential for mediating mitochondrial fusion. It shapes mitochondrial networks, sustains oxidative phosphorylation efficiency, and modulates cell apoptosis via interactions with MFN1. Additionally, MFN2 tethers mitochondria to the endoplasmic reticulum, facilitating calcium signaling and lipid exchange.
Mutations in MFN2 cause Charcot-Marie-Tooth disease type 2A2 (CMT2A2), a peripheral neuropathy. Dysregulation links to insulin resistance, Parkinson’s disease, and heart failure. Preclinical studies focus on small molecules targeting MFN2 to restore fusion; candidates show promise in ameliorating metabolic and neurodegenerative phenotypes, though no approved therapies exist currently.

IHC image of CSB-RA799368A0HU diluted at 1:100 and staining in paraffin-embedded human kidney tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
