Mucin 4 (MUC4) is a transmembrane glycoprotein consisting of α and β subunits. Its β subunit binds to ErbB2 (HER2), activating PI3K/AKT and MAPK signaling pathways to drive cell proliferation, migration, and resistance to apoptosis. Common aliases include MUC-4 and transmembrane mucin 4.
Overexpressed in pancreatic cancer, MUC4 acts as a diagnostic biomarker and prognostic factor; it’s also associated with lung and breast cancers. Drug development targets MUC4-ErbB2 interaction: monoclonal antibodies and small-molecule inhibitors are in preclinical stages, showing efficacy in suppressing tumor growth in vitro and animal models.
