NKX3-2

NKX3-2 (NK3 homeobox 2), also known as BAPX1, is a NK homeobox family transcription factor critical for chondrogenesis. It regulates chondrocyte differentiation and maturation via modulation of TGF-β/BMP signaling pathways, supporting axial/appendicular skeleton development and temporomandibular joint formation.
Loss-of-function mutations in NKX3-2 cause spondyloenchondrodysplasia, a rare skeletal disorder. Its downregulation in colorectal cancer implies a tumor suppressor role. While direct therapeutics are preclinical, its role in cartilage homeostasis positions it as a target for regenerative treatments for osteoarthritis and cartilage injuries.
