OGFOD1, full name 2-oxoglutarate and iron dependent oxygenase domain containing 1, has aliases TPA1, KIAA1612 and FLJ10826. As a 2OG-Fe(II) dioxygenase, it catalyzes RPS23 prolyl hydroxylation to maintain translational fidelity, modulates eIF2α phosphorylation and stress granule dynamics. It regulates iron metabolism and AMPK/mTOR pathways, and mediates ischemic signaling by regulating ATPAF1 expression and SPARC translation.
OGFOD1 dysregulation links to hepatic ischemia-reperfusion injury and myocardial ischemic damage, and its overexpression accelerates stress-induced apoptosis. Preclinically, selective OGFOD1 prolyl hydroxylase inhibitors have been developed via structural design. Silencing OGFOD1 is a promising strategy for alleviating oxidative stress in liver injury, with its modulators under experimental research for ischemic disease therapy.
