OTUD1 encodes OTU domain-containing protein 1, a deubiquitinating enzyme (DUB) from the OTU superfamily. It catalyzes ubiquitin chain removal from substrate proteins, regulating their stability or activity. Key pathways include DNA damage response (modulating RNF168) and NF-κB signaling (targeting TRAF6).
Dysregulation of OTUD1 is linked to breast/lung cancers via its DDR and proliferation roles; emerging ties to neurodegeneration exist. Preclinical efforts focus on selective small-molecule inhibitors of its DUB activity—no approved therapies yet, but these agents show promise for treating OTUD1-associated disorders.
