PRSS1

PRSS1 encodes Protease Serine 1, also called Trypsinogen-1—a pancreatic zymogen central to protein digestion. Activated to trypsin by enteropeptidase in the small intestine, it triggers proteolysis and activates other digestive enzymes like chymotrypsinogen.
Gain-of-function PRSS1 mutations cause hereditary pancreatitis, as premature trypsin activation in the pancreas induces inflammation. Current drug development focuses on serine protease inhibitors targeting trypsin activity; several preclinical candidates show potential in mitigating pancreatic damage from excessive trypsin activation.
