PSMD11

PSMD11 encodes 26S proteasome non-ATPase regulatory subunit 11 (alias RPN6), a core component of the 26S proteasome’s regulatory particle. It mediates ubiquitinated protein degradation via the ubiquitin-proteasome system (UPS), maintaining intracellular protein homeostasis. It also modulates NF-κB signaling by facilitating IκBα degradation, enabling NF-κB activation and downstream pro-inflammatory or oncogenic responses.
Dysregulation of PSMD11 is linked to cancers (e.g., lung, breast) due to impaired UPS promoting oncoprotein accumulation. It correlates with neurodegenerative diseases like Alzheimer’s, where UPS dysfunction leads to toxic protein aggregates. Current research explores PSMD11 as a prognostic biomarker and develops small molecules targeting its activity to restore UPS function in pathological states.
