It recruits DNA polymerase η to stalled replication forks via monoubiquitination of PCNA, facilitating translesion synthesis. RAD18 interacts with pathways like homologous recombination and nucleotide excision repair, maintaining genomic stability.
Dysregulation of RAD18 is linked to cancer development, particularly breast and colorectal cancers, due to impaired DNA repair. Preclinical studies explore small-molecule inhibitors targeting its ubiquitin ligase activity, aiming to enhance chemotherapy sensitivity. However, clinical translation remains in early stages.
