SHMT1

Serine Hydroxymethyltransferase 1 (SHMT1), also called cytosolic SHMT (cSHMT), catalyzes serine and tetrahydrofolate conversion to glycine and 5,10-methylenetetrahydrofolate. A core enzyme in one-carbon metabolism, it fuels purine/thymidylate synthesis and methyl transfer—key for cell proliferation and epigenetic regulation.
SHMT1 variants link to neural tube defects; its overexpression in cancers like colorectal carcinoma promotes tumor growth. Preclinical inhibitors (e.g., SHIN1) target SHMT1 to disrupt thymidylate production, enhancing fluorouracil’s chemotherapeutic efficacy by blocking cancer cell nucleotide synthesis pathways.
