SLC1A5, also known as Alanine-Serine-Cysteine Transporter 2 (ASCT2), is a sodium-dependent neutral amino acid transporter primarily responsible for glutamine uptake. It belongs to the solute carrier family 1 and functions as a key regulator of cellular metabolism by facilitating the transport of glutamine, alanine, serine, and cysteine. ASCT2-mediated glutamine import supports mTORC1 signaling and c-Myc-driven metabolic reprogramming, critical pathways in cancer cell proliferation and survival. Overexpression of SLC1A5 is observed in multiple malignancies, including colorectal, lung, and triple-negative breast cancers, where it correlates with poor prognosis and chemoresistance. Recent studies also implicate its role in neurological disorders such as Alzheimer's disease through disrupted glutamate-glutamine cycling. Pharmacological inhibitors like V-9302 and benzylserine derivatives are under preclinical investigation, targeting ASCT2 to starve tumors of glutamine or modulate neurotransmitter balance.
