TBC1D31, a member of the TBC1 domain family, is expressed abundantly in human cells with particular prominence in testis and lymph nodes. This protein functions as an activator of Rab GTPase and is implicated in the downregulation of RabB22A-mediated intralysosomal transport and EGFR degradation by promoting GTP hydrolysis of RabB22A, which in turn activates EGFR signaling. TBC1D31 plays a pivotal role in hepatocellular carcinoma, indicating its potential as a novel therapeutic target for patients with genetic amplification or overexpression of the TBC1D31 gene.
