TFR2, a member of the transferrin receptor-like family, is a single transmembrane protein predominantly found in liver and intestinal cells. It regulates the cellular iron uptake by binding to transferrin, thereby influencing iron metabolism, liver cell function, and the differentiation of red blood cells. Unlike other iron-regulated proteins, TFR2's expression is not modulated by intracellular iron levels. It is notably expressed in liver cells, erythroid precursor cells, K562 cells, and HepG2 cells. Mutations in TFR2 are implicated in hereditary hemochromatosis type III, underscoring its critical role in iron homeostasis. Furthermore, TFR2 contributes to the differentiation and development of erythroid precursor cells and serves as a molecular marker for these cells, highlighting its multifaceted impact on erythropoiesis and iron regulation.

IHC image of CSB-PA14729A0Rb diluted at 1:200 and staining in paraffin-embedded human liver cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of MCF-7 cells with CSB-PA14729A0Rb at 1:66, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
