TGDS, full name TDP-glucose 4,6-dehydratase, has major aliases SDR2E1, TDPGD and CATMANS. As a member of the SDR superfamily with a NAD-binding domain, it catalyzes TDP-glucose dehydration to generate UDP-4-keto-6-deoxyglucose, a metabolite rescuing UXS1 activity. It modulates nucleotide sugar metabolism and glycosaminoglycan synthesis, and its enzymatic activity is essential for maintaining the TGDS-UXS1 metabolic axis.
TGDS loss-of-function mutations cause Catel-Manzke syndrome with craniofacial and skeletal anomalies, and its dysregulation links to abnormal nucleotide sugar metabolism. No clinical drugs target TGDS yet; preclinical research focuses on supplementing its catalytic metabolite to restore UXS1 activity and glycosaminoglycan synthesis, providing a promising direction for treating TGDS-related rare skeletal dysplasias.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. Secondary antibody only control: uses 1% BSA instead of primary antibody
