Transmembrane Protein 33 (TMEM33), also known as FLJ10213, is an endoplasmic reticulum (ER)-resident transmembrane protein. It modulates calcium homeostasis by interacting with inositol 1,4,5-trisphosphate receptors (IP3Rs), regulating ER calcium release. This function links it to the unfolded protein response (UPR) pathway, where it helps mitigate ER stress by fine-tuning calcium signaling dynamics.
TMEM33 dysregulation is associated with neurodegenerative diseases (e.g., Alzheimer’s) due to impaired ER stress resolution, and cardiovascular disorders from calcium signaling disruptions. While no targeted therapies are approved yet, preclinical research is investigating small molecules targeting TMEM33-IP3R interactions to restore ER homeostasis in disease models.
