USP14

USP14 is a deubiquitinating enzyme belonging to the ubiquitin-specific protease family, also known as UBP6 in yeast homologs. It regulates protein degradation by associating with the 26S proteasome, where it counteracts ubiquitin chain trimming to modulate substrate access and proteasomal activity.
USP14 has been linked to multiple signaling pathways, including NF-κB and mTOR, and its dysregulation is implicated in cancer, neurodegenerative diseases like Alzheimer’s, and viral infections. Small-molecule inhibitors targeting USP14, such as IU1 and its analogs, are under preclinical investigation for enhancing proteasomal degradation of misfolded proteins and treating malignancies.
